Представить методы изучения портальной гипертензии и способы ее медикаментозной коррекции в эксперименте.Основные положения. Для изучения сложных патофизиологических нарушений, свойственных портальной гипертензии, используются различные экспериментальные модели, которые позволяют дать им комплексную оценку и разработать способы адекватной коррекции. В настоящее время на лабораторных животных детализированы механизмы развития гипердинамического циркуляторного статуса, исследования ex vivo дали физиологическую характеристику медиаторам, вызывающим спланхническую вазодилатацию и увеличение сосудистого тонуса в цирротически измененной печени, а методы молекулярной биологии идентифицировали дефекты в сигнализирующих путях, ответственных за нарушение метаболизма этих веществ.Заключение. В последние годы применение различных экспериментальных моделей портальной гипертензии способствовало прогрессу не только в понимании лежащих в ее основе патофизиологических механизмов, но и разработке новых перспективных методов лечения. Дальнейшие рандомизированные контролируемые клинические испытания дадут возможность определить их безопасность и эффективность при сравнении с традиционными способами терапии.</p></abstract><trans-abstract xml:lang="en"><sec><title>The aim of review</title><p>The aim of review. To present methods of studying of portal hypertension and means of its pharmacological treatment in experiment settings.</p></sec><sec><title>Original positions</title><p>Original positions. To study complex pathophysiological disorders, innate to portal hypertension, various pilot models providing complex evaluation and development of adequate treatment methods are used. Nowadays mechanisms of hyperdynamic circulatory status development are detailed on laboratory animals. Ex vivo studies gave physiological characteristic to the mediators causing splanchnic vasodilation and increasing of vascular tension in cirrhotic liver, and methods of molecular biology identified defects in signalling routes responsible for disorders of metabolism of these substances.</p></sec><sec><title>Conclusion</title><p>Conclusion. In the last years application of various pilot models of portal hypertension promoted progress not only in understanding of pathophysiological mechanisms laying in its basis, but also development of new perspective methods of treatment.
